Mechanism of Action
Unclear; possible inhibition of DNA, RNA and protein synthesis, and alkylation of DNA. Cell cycle-specific with greatest activity during S phase.
Indications
Hodgkin’s disease, brain tumors, multiple myeloma, lym-phomas, small cell lung cancer.
Metabolism/Excretion
Metabolized in the liver; excreted in urine.
Dosage Range
♦ Adult: Initially, 2 to 4 mg/kg per day PO for 7 days, then 4 to 6 mg/kg per day until maximum response achieved; maintenance, 1 to 2 mg/kg per day.
