Mechanism of Action

Cell cycle-nonspecific agent that interacts with DNA and inhibits the DNA enzyme topoisomerase, causing cell lysis.


Indications

Acute monocytic leukemia, acute myelocytic leukemia, acute promyelocytic leukemia.


Metabolism/Excretion

Metabolized in the liver; primarily excreted in urine and feces.


Dosage Range

♦ Adult: 12 mg/m2 per day for 2 to 3 days in combination therapy with cytosine arabinoside 100 mg/m2 continuous infusion for 5 to 7 days. Cumulative cardiac dose is 120 to 140 mg/m2, depending on history of mediastinal radiation or anthracycline therapy.
