Mechanism of Action

Cell cycle-nonspecific alkylating agent that alkylates and impairs DNA replication, causing cell lysis.


Indications

Ovarian carcinoma; multiple myeloma; bone marrow transplant cytoreduction for neuroblastoma, melanoma, lymphoma, myeloma, and colon cancer.


Metabolism/Excretion

Metabolized by hydrolysis in plasma; excreted in urine and feces.


Dosage Range


Adult

♦ Ovarian carcinoma: 0.2 mg/kg per day for 5 days, repeated every 4 to 5 weeks

♦ Multiple myeloma: 6 mg/day for 2 weeks; subsequent doses depend on recovery from previous course. IV dose: 16 mg/m2 every 3 weeks for four doses, then every 4 weeks.
