Mechanism of Action

Essential for purine and DNA synthesis; bypasses the inhibitor action of folic acid antagonists.

Indications

♦ Folinic acid rescue after high-dose methotrexate for osteosarcoma, epidermoid tumors, and various refractory tumors

♦ Megaloblastic anemias due to nutritional deficiency, pregnancy, infancy, and sprue

♦ Palliative treatment with 5-fluorouracil for metastatic colorectal cancer

Metabolism/Excretion

Metabolized in the liver; primarily excreted in urine, small amounts in feces.
