Mechanism of Action
Docetaxel belongs to the taxoid family and acts by disrupting the microtubular network in the cells, which is essential for their mitotic and interphase functions.
Indications
Locally advanced or metastatic breast carcinoma after progression of the disease with anthracycline-based therapy or relapse during anthracycline-based adjuvant treatment.
Metabolism/Excretion
The oxidative metabolites, which are metabolized in the liver, are excreted in the urine and feces, mainly in the feces. Half-life: 11 hours.
Dosage Range
♦ Adult: 60 to 100 mg/m2 IV every 3 weeks. Patients who are initially given 100 mg/mL and experience neu-tropenic fevers, a neutrophil count of <500 cells/mm3 for more than 1 week, severe or cumulative cutaneous reactions, or severe peripheral neuropathy should have the dose reduced to 75 mg/m2. If symptoms persist at the new
level, the dose may be further reduced to 50 mg/m2 or discontinued. Patients who are started at a lower dose of 60 mg/m2 and who do not experience the above symptoms may be given higher doses.
